Gentamicin derivs. I and II were prepd. from 6'-N-tert-butoxycarbonylgentamicin B by
acylation with N-(S-4-benzyloxycarbonylamino-2-hydroxybutyryloxy)- and
N-(S-3-benzoyloxycarbonylamino-2-hydroxypropionyloxy)succinimide, followed by
removal of the benzyloxycarbonyl group by catalytic hydrogenation and removal of the
tert-butoxycarbonyl group with CF3CO2H. 3-N-isomer of I was also formed in a small amt.
In vitro antimicrobial activities of I and II against a variety of
gentamicin sensitive strains, adenylating strains, and
acetylating strains of microorganisms are compared with
gentamicin, tobramycin, and amikacin.
